Using the HiLo LUT in ImageJ, raw data and intensity measurements.
The point is that saturatio is not required to get good images and quantifiable images.
The next picture shows the same images with gamma 0.58 applied. Whereas the intensity measurements are not valid for assessing the biology, the images are valid for presentation to show the deatils inthe dark regions as long as the methods clearly describe the gamma adjustment.
The next picture down, which is raw data taken with the gain turned up, is no improvement and is less pleasing by eye because it appears noisier.
