To test the PicoQuant system on the Zeiss 880 confocal in June 2016 a postdoc prepared cells expressing CFP alone and cells expressing CFP-YFP separated by 6 bases thinking this would result in FRET.
Based on these results we believe we imaged Forster Resonance Energy Transfer in the time domain using the PicoQuant pulsed 445 nm laser as excitation source and emission in 460-500 nm range.
We are new at the Symphotime software. We simply drew ROIs on the images and did single exponential fits. The full results window looks like this and in the figures below we report the number in the yellow ellipse. These are encouraging preliminary data.
