The point of this webpage is to show a quick local contrast enhancement method to pull out spatial details. Really only valid when know objects of interest are puncta.

For sCMOS camera, a flatfield correction step to remove single pixel noise based on the camera chip may be added.
In ImageJ 1.51 or later run this macro which should work on any multichannel images; no error checking to confirm images valid for the macro. puncta_cleanup_macros_v100.txt
Additional 1x1 median flitering:

What this macro doesn't do, which a future version will do, is combine Z sections of the same channels. The following example is three serial sections of each green and red through time.
