Each square is 10 X 10 um. Thorlabs calibration slide.

If you have the correct lens set in Nikon Elements software and you save the files as .nd2, then importing into ImageJ using bioformats_package.jar at http://www.openmicroscopy.org/site/support/bio-formats5.1/users/imagej/ will automatically pull in the spatial scale information. If you have saved the files as .tif or .jpg (the latter is just plain wrong), then you need to set the scale info below manually.

20X LWD lens 0.33 um/pixel	40X air lens 0.16 um/pixel
